Notch

Notch

Notch signaling functions in a wide range of cell types and at different stages of development and is evolutionarily conserved. Transmembrane ligands on one cell activate transmembrane receptors on a cell that is next to it, which is how notch signaling takes place.
For the majority of eukaryotic organisms to develop and remain healthy, Notch signaling regulation is essential. Direct cell-cell interactions are required to activate the Notch signaling, which is mediated by the Notch receptors (NOTCH1, 2, 3, and 4) and ligands (DLL1, 3, and 4, JAG1 and 2), which are essential membrane proteins. Through an unusual mechanism involving Notch proteolysis to release the intracellular domain from the membrane, ligand-expressing cells activate Notch signaling. This enables the Notch receptor to directly serve as the downstream signal transducer.
