VHL 7 nM (EC50) SMARCA2
ln Vitro
In RKO cells, ACBI2 breaks down SMARCA2 and SMARCA4 with DC50s of 1 nM and 32 nM, respectively [1]. SMARCA2 is rapidly and fully degraded in two sensitive cell lines (A549 and NCI-H1568) by ACBI2 (0.1 nM-1 μM, 4–18 hours)[1]. Compared to SMARCA4, ACBI2 (1 nM-1 μM, 18 hours) can more selectively and significantly degrade SMARCA2[1]. In human whole blood, ACBI2 (0-1 μM; 18 hours) destroys SMARCA2 in a dose-dependent manner [1].
ln Vivo
In the A549 xenograft mouse model, ACBI2 (80 mg/kg, orally, once day) effectively suppresses tumor growth [1]. In A549-transplanted tumor-bearing mice, ACBI2 (5-100 mg/kg, orally, tumors taken 24 or 48 hours after treatment) dose-dependently destroys tumor SMARCA2 (IHC staining) [1]. Of ACBI2 (30 mg/kg, oral, mouse), 22% is bioavailable orally [1].
