Guidelines (This is our suggested protocol; it should be adjusted based on your unique requirements as it just offers as a guideline). A. Cell Labeling: 1. Proceed with the regular protocol for incubating the cells. 2. The cells are kept in the dark at 37°C in a CO2 incubator with DMEM containing 5 μM TBR. 3. After an hour, switch to regular culture medium without phenol red. The cells were then exposed to light using a 500 W Xe arc that was filtered. B. The location of TBR within cells: 1. For 48 hours, cells are cultivated in glass dishes with a 35 mm diameter. Proceed with the regular protocol for incubating the cells. 2. To perform triple staining, switch the medium to a solution that contains 5 μM of BODIPY-TR ceramide. 3. After incubating the cells for 20 minutes at 48°C, they are then incubated for an additional hour at 37°C. Following this, the solution is replaced to regular media containing 5 μM TBR. 4. 5. The cells were incubated for 15 minutes after the media (final concentration, 100 μM) was supplemented with the stock solution (1 mM) of Hoechst 33342. Following a wash with phosphate-buffered saline (containing Ca2+ and Mg2+), cells are examined using confocal laser scanning microscopy (LSM).
