An agonist, nelonicline citrate, binds to human brain α7 nAChRs with high affinity (Ki = 12.3 nM) and activates currents in Xenopus oocytes that express human α7 nAChRs that have been recombinant (EC50 = 2 μM; 74% intrinsic activity relative to acetylcholine). In human IMR-32 neuroblastoma cells, citrate does bind to α3β4* nAChRs (Ki=60 nM), however in a calcium flux assay, it is only 12% effective at 100,000 nM. Nelonicline citrate is a 5-HT3 receptor antagonist, just like certain other α7 nAChR agonists, but its affinity for this receptor is >10-fold lower than that of α7 nAChRs (Ki of 140 nM)[1].
