When exposed to 37˚C and pH 7.4, pozelimab binds to both human and cynomolgus C5, with Kd values of 262 pM and 8.55 nM, respectively [1]. In human and cynomolgus monkey serum, pozelimab inhibits representative pathway (CP)-mediated hemolysis with IC50 values of 2.8 and 20 nM, respectively, at 37˚C, pH 7.4 [1]. In serum from normal and cynomolgus monkeys, pozelimab blocks alternative pathway (AP)-mediated hemolysis with IC50 values of 26 and 8.9 nM, respectively, at 37˚C and pH 7.4 [1]. Human C5, cynomolgus C5, human R885C C5, and human C5b,6 are all bound by pazelimab at pH 7.4 and 25˚C, with Kd values of 0.189, 2.73, 0.422, 0.465, and 0.137 nM, respectively [1].
