PROTAC EED degrader-2 (compound PROTAC 1) has a pKD of 9.27±0.05 for binding to EED and a pIC50 of 8.11±0.09 for inhibiting PRC2 function [1]. The EZH2 mutant DLBCL cell line Karpas422 can be successfully inhibited from proliferating by PROTAC EED degrader-2 (0.01-100 μM; 14 days) [1]. In Karpas422 cells, PROTAC EED degrader-2 (0.1-3 μM; 48 hours) lowers the protein levels of EED, EZH2, and H3K27me3 [1]. In Karpas422 cells, PROTAC EED degrader-2 (1 μM; 1-24 h) stimulates the breakdown of EED, EZH2, and SUZ12 [1]. Reduced proliferation was the outcome of the cell proliferation assay [1] using Karpas422 cells as the cell line. Concentration: 0.01–0.1, 1, 10, 100 μM. Incubation time: 4, 8, 12, and 14 days. GI50 on Day 14 is 0.057 μM.

Solubility in Formulation 1: ≥ 2 mg/mL (2.08 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution. Solubility in Formulation 2: ≥ 2 mg/mL (2.08 mM) (saturation unknown) in 10% DMSO + 90% Corn Oil (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.0 mg/mL clear DMSO stock solution to 900 μL of corn oil and mix evenly.  (Please use freshly prepared in vivo formulations for optimal results.) |
