NADPH-oxidase (NOX) is a highly regulated dynamic complex comprising membrane and cytosolic proteins and is the major source of nonmitochondrial cellular reactive oxygen species (ROS). The redox-dependent signaling cascades, innate immunity, hormone production, and other physiological processes depend on the ROS produced by NADPH oxidases. They also act as cofactors in these processes.
The seven members of the NOX enzyme family of transmembrane proteins—NOX1 through NOX5, as well as DUOX1 and DUOX2—each have a unique tissue distribution and activation mechanism. They facilitate the conversion of molecular oxygen into superoxide anion, which then catalyzes the rapid reaction that produces other ROS, including hydrogen peroxide. Although minimal NOX activity is essential for healthy physiology, NOX enzyme overactivity results in disease. NADPH oxidases are becoming more and more popular as drug targets.
