Leukotrienes, lipoxins, and neuroprotectins are just a few of the eicosanoids and docosanoids that are produced as a result of the metabolism of arachidonic and docosahexaenoic acid by the family of enzymes known as lipoxygenases (LOXs). These enzymes are either involved in oxidative stress and cell death mechanisms or in cell protection, depending on the redox state of the cells and other milieu conditions. Lipoxygenases are lipid peroxidizing enzymes that may be targets for pharmacological treatment because they are involved in the pathogenesis of inflammatory and hyperproliferative diseases.
According to where arachidonate is oxygenated, lipoxygenases are categorized as 5-, 8-, 9-, 11-, 12-, and 15-LOX. 5-LOX, 8-LOX, 12-LOX, and 15-LOX are the most prevalent animal LOXs, whereas 5-LOX and 15-LOX are the most prevalent plant LOXs. When it comes to the production of 5-hydroperoxyeicosatetraenoic acid (5-HpETE) and other bioactive lipid mediators, 5-LOX is the most prevalent isoform.
