PROTAC Linkers

PROTACs (Proteolysis Targeting Chimeric Molecules) are heterobifunctional protein degraders and promising targeted therapeutics candidates for cancer.Target protein binders and E3 ligase recruiters, two functional motifs of a PROTAC, are joined by the PROTAC linker.

A PROTAC's linker has a crucial function. The characteristics of the linker (type, length, attachment position) can influence how ligase is formed. The PROTAC:target ternary complex affects the target protein's effective ubiquitination and eventual destruction. According to reports, the ideal linker lengths for PROTACs range from 12 to over 20 carbons. PEGs, alkyl chains, and alkyl ether are the most often employed linkers in the creation of PROTACs.
