THPP-1 is a novel, potent and orally bioavailable PDE10A (phosphodiesterase 10A) inhibitor with Ki values of 1 nM and 1.3 nM for human and rat PDE10A, respectively.

Rats given THPP-1 (0.1–1.0 mg/kg) orally show unique item identification skills [1]. In the rat striatum, THPP-1 (oral, 1-30 mg/kg) dose-dependently raises GluR1 S845 phosphorylation while having no effect on total GluR1 levels in the striatum. At every dose looked at in this investigation, the administration of THPP-1 also led to an increase in the pGluR1/GluR1 ratio [1]. The effects of THPP-1 (1, 3, and 10 mg/kg) orally on monkeys' stimulant-induced locomotor activity (SIMA) were dramatically reduced [2].
