Angiotensin 1-7 (Ang-(1-7)) is an endogenous heptapeptide in the renin-angiotensin system (RAS) due to its anti~inflammatory and anti-fibrotic activities in cardiomyocytes Cardioprotective effect. Angiotensin 1-7 inhibits the activity of purified canine ACE (angiotensin-converting enzyme) with IC50 of 0.65 μM. Angiotensin 1-7 works as a local coregulator of kinin-induced vasodilation by inhibiting ACE (angiotensin-converting enzyme) and releasing nitric oxide. Angiotensin 1-7 blocks Ang II-induced smooth muscle cell growth/proliferation and hypertrophy and displays anti-angiogenic and growth-inhibitory activities on endothelium. Angiotensin 1-7 displays anti~inflammatory activity.
Biological Activity I Assay Protocols (From Reference)
ln Vitro
The growth of cultured vascular smooth muscle cells is inhibited by angiotensin 1-7 (Ang-(1-7)), while the proliferation of cells is stimulated by equimolar doses of Ang II [2]. In NRK-52E cells, angiotensin 1-7 (Ang 1-7) prevents the persistent activation of the TGF-β-ERK pathway, hence eliminating the phenotype of myofibroblasts activated by methylglyoxal-modified albumin (MGA) [4]. In opposition to the Ang II/angiotensin II type 1 receptor (AT1R), angiotensin 1-7 signals through the Mas receptor (MasR), promoting anti-inflammatory, vasodilatory, and neuroprotective actions [5].
ln Vivo
The treatment of DSS-induced colitis with daily angiotensin 1-7 (Ang-(1-7)) at 0.01-0.06 mg/kg was dramatically improved. The phosphorylation of p38, ERK1/2, and Akt associated with colitis is decreased by Ang 1-7 therapy [3].
