The class B G-protein coupled family of receptors includes the glucagon receptor, a 62 kDa protein that is activated by glucagon and coupled to G alpha i, Gs, and to a lesser extent G alpha q. Adenylate cyclase is activated and intracellular cAMP levels are raised when the receptor is stimulated. The GCGR gene in humans is responsible for encoding the glucagon receptor. The liver and kidneys are the primary sites of glucagon receptor expression, with smaller amounts of these receptors also present in the heart, adipose tissue, spleen, thymus, adrenal glands, pancreas, cerebral cortex, and gastrointestinal tract.
